CB-03-01, why not just make it?

Hey all. I just joined this community a few minutes ago. This is my first post. Straight up, I'm looking for the organic chemist amongst you all, if one exists. I am medical student in the US (almost Dr. lottasoda). I'd like to examine the feasibility of manufacturing CB-03-01 for personal use, but that process is a little outside my realm of expertise. Thus, anyone who knows how to esterify 11-deoxycortisol...lol, and where to get it, please let me know.

I tried to read the other posts on CB-03-01. I'm only interested in this "project" b/c it sounds like the Chinese supplier is not totally legit or cost effective. You guys collectively are very thorough in your discussions, but there's a lot of volume to get through. If I've missed some solution on the whole CB-03-01 availability issue, please let me know

Thanks

We don't have a vehicle (other than maybe injecting it), forget it for now. It's still in trials, phase II I believe. IMO it's the best option we have in the pipeline.

I agree that it's a good looking option. I don't know about an injection, but perhaps the microdermal rollers could come into play here. Phase II means they are double checking the safety, and trying to demonstrate efficacy. This step is completed for the acne trials. CB-03-01 checks out for facial acne, the only difference when it comes to Androgenetic Alopecia is application to the scalp. Dosing doesn't change, right? The scalp has a more significant circulation, so systemic absorption of the drug may increase, i.e. more "side effects".

Moreover, if it's going to phase III trials with the manufacturer, then a vehicle exists. It's essentially a fairly lipophilic looking steroid hormone. I can't imagine the vehicle is the hard part in making one's own product for personal use.

I'll talk to some of the pharmacists at the hospitals about how to approach the vehicle problem...

If the application site is small, you could just learn how to properly inject with a syringe! But if you are serious about using this as a long term component to your regimen, and the application site is large, get a mesotherapy gun and make micro injections, it's fool proof! They used iontophoreris in the trials with success.

I really want to know what vehicle they plan to use, It is a big *** hydrophobic molecule! It will be hard to get it past the dermis and to the hair follicles.

As far as sides go, I feel it is pretty safe to inject, based on the fact they administered it via iontophoresis, and, even though we are not rats lol, this is encouraging...

"CB-03-01, like its parent cortexolone, was ineffective
as antiandrogen when subcutaneously administered to
rats up to a daily dose equivalent about to 100 mg/kg.
The observed dissociation between the strong topical
antriandrogenic activity and the absence of systemic effect
could be explained supposing that, after subcutaneous
injection, CB-03-01 undergoes a fast hydrolysis,
by the liver or other tissues, leading to the inactive cortexolone
form [12]. Indeed, in our assays, the parent
cortexolone was devoid of both topical and systemic
antiandrogenic effects, thus indicating that CB-03-01
acts “per se” and not as a prodrug. Considering that
finasteride and flutamide were reported to induce a significant
inhibition of hamster’s sexual accessory organs
after topical application of low dose of about 0.25 mg/
kg [13], and that bicalutamide displayed similar effects
in rat after oral administration of 0.4 mg/kg [14], our
data are very impressive and clearly indicate that the
topical administration of CB-03-01 should not have systemic
effects, on androgen-sensitive organs and functions,
even in case of large percutaneous absorption."

Looks like "no systemic effects on androgen-sensitive organs" is their biggest selling point...

But what about those non sex-related sides? Like fatigue and sleep problems. I don't really think they happen just because AA drugs mess with your sexual organs.

And seriously cortisol plays big roles in your body
